Adapting a published method to your reagents and equipment

A published method is written for their bench: their kit, their rotor, their buffer stocks, their sample amounts. The model cannot see yours, so an adaptation is only as good as the inventory you hand it - which reagents you actually have, what your centrifuge tops out at, what your starting material is. Given that context, the useful output is a draft protocol with every departure from the published version flagged as a DEVIATION, each with a note on what it might change. What it is not: a validated method. The model drafts the adaptation and lists what a pilot run must confirm; whether the adaptation works is something the pilot decides, not the text.
